硫酸葡聚糖钠盐(Dextran sulfate,DSS)是葡聚糖的聚阴离子衍生物,由葡聚糖和sulfurochloridic acid的酯化反应形成。其中含硫量 约为 17%,相当于葡聚糖分子的每个葡萄糖残糖中平均含1.9个硫酸基团。
DSS具有几个特点:
1)聚阴离子复合物,可溶 于水,形成无色水溶液;
2)纯度高,且具有良好的稳定性;
3)可被自然降解。
炎症性肠炎(IBD)是一种慢性、易复发的胃肠道感染,会提升肠道肿瘤发生的危险性,主要包括 UC 和克罗恩病(Crohn disease, CD)。自 1985年首次报道采用葡聚糖硫酸钠(dextran sulphate sodium, DSS)制备出仓鼠溃疡性结肠炎模型以来,已有大量研究证明 DSS 结肠炎模型与人类溃疡性结肠炎相似。DSS结肠炎模型的组织学特点、临床表现、发病部位和细胞因子增殖情况都与人类溃疡性结肠炎(ulcerative colitis, UC)极为相似。该模型的造模条件和操作方法简单,造价便宜,重复性好,便于掌握和推广;可根据实验目的调整 DSS 浓度和给药时间,建立急性、慢性和急慢性交替性模型。
